Breast adenocarcinoma MCF7

cell line / Homo sapiens / breast adenocarcinoma

How to order?
Animal species:
Human, Homo sapiens.
Origin:
metastatic breast adenocarcinoma
Age:
elderly
Tissue:
mammary gland
Morphology:
epithelial, adherent
Histological diagnosis:
breast adenocarcinoma
Tumoroids obtained:
yes
Subcutaneous growth:
yes
Molecular profile:
wnt7b oncogene, Blood group I(O) antigen. Rh factor, +, insulin-like growth factor binding protein (IGFBP) BP-2, 4 and 5. Markers Estrogen receptor, Isoenzymes AK-1, ES-D, G6PD, GLO-I, Me-2, PGM1, PGM3
Tumorigenicity:
yes, tumors in nude mice
Karyology:
modal number = 82. range = 66 to 87. The stemline chromosome number varied from hypertriploid to hypotetraploid, with the 2S component occurring in 1% of cases. There were 29 to 34 marker chromosomes per S-metaphase. 24 to 28 markers were present in at least 30% of cells, and typically one large submetacentric (M1) and 3 large subtelocentric (M2, M3 and M4) markers were recognized in more than 80% of metaphases. No SD were found. Chromosome 20 was nullisomic and X was disomic.
Contamination control:
not detected
Viral infection:
not performed
Species identity control:
karyological, isoenzyme (LDH and G6PD) and immunofluorescence analysis.
Application area:
3D cell culture, Immuno-oncology
Culture method:
in vitro partially adhesion, in vivo (see Appendix 1).
In vitro culture conditions:
medium – EMEM fetal bovine serum 10%. passaging procedure - cell detachment using trypsin 0.25 %: Versene 0.02 % (1:3), split ratio 1:3 -1:5 cryopreservation – growth medium 65%, 5% DMSO, 30% fetal bovine serum Slowly growing cell line and poorly attached three-dimensional clusters with some floating viable cells. They should be saved by gentle centrifugation (125 × g for 5-10 minutes) and added back. Increasing the serum concentration to 20% may also help improve growth. Human recombinant insulin can be used instead of bovine insulin when preparing complete medium for MCF7(see Appendix 1).
Physical state of the sample:
frozen.
Cryopreservation:
liquid phase and nitrogen vapor (see Appendix 1).
Viability after cryopreservation:
94% (trypan blue staining after thawing).
Collections:
A.P. Avtsyn Research Institute of Human Morphology, Federal State Budgetary Scientific Institution "B.V. Petrovsky Russian Scientific Center of Surgery", ATCC.
Patent, certificate:
no
Note:
transfection is possible. Metastasis as pleural effusion retains several characteristics of differentiated mammary epithelium, including the ability to process estradiol via cytoplasmic estrogen receptors and the ability to form domes. MCF7 cell growth is inhibited by tumor necrosis factor alpha (TNF alpha). IGFBP secretion can be modulated by treatment with antiestrogens.
Intended use:
these samples are intended for laboratory research only.
