Tumor induced by Abelson murine leukemia virus RAW 264.7
cell line / Mus musculus / monocytic leukemia
- Animal species:
- Mouse Mus musculus.
- Origin:
- monocytic leukemia
- Age:
- sexually mature
- Tissue:
- ascites
- Morphology:
- monocytes/macrophages
- Histological diagnosis:
- monocytic leukemia
- Tumoroids obtained:
- yes
- Subcutaneous growth:
- yes
- Molecular profile:
- Genes expressing lysozyme. H-2d.
- Tumorigenicity:
- yes, tumors in BALB/c mice
- Contamination control:
- not detected
- Viral infection:
- ectromelia virus (mousepox) negative.
- Species identity control:
- karyological, isoenzyme (LDH and G6PD) and immunofluorescence analysis.
- Application area:
- oncology, immunology
- Culture method:
- in vitro adhesion, colonies in soft agar, in vivo (see Appendix 1).
- In vitro culture conditions:
- Medium – DMEM fetal bovine serum 10%. passaging procedure - cell detachment using trypsin 0.25 %: Versene 0.02 % (1:3), split ratio 1:3 -1:5 cryopreservation – growth medium 65%, 5% DMSO, 30% fetal bovine serum Viable cells may detach and float, especially as the culture becomes more homogeneous. These cells are still viable and should be saved by gentle centrifugation. Leave 10 mL of medium in the flask before scraping. When using a scraper, work carefully to make only one pass through the flask, from bottom to top, to avoid damaging the cells. Viability usually decreases after subculturing by scraping, and the cells may take longer to attach to the flasks. (see Appendix 1).
- Physical state of the sample:
- frozen.
- Cryopreservation:
- liquid phase and nitrogen vapor (see Appendix 1).
- Viability after cryopreservation:
- 94% (trypan blue staining after thawing).
- History:
- obtained in 1978
- Collections:
- A.P. Avtsyn Research Institute of Human Morphology, Federal State Budgetary Scientific Institution "B.V. Petrovsky Russian Scientific Center of Surgery", ATCC.
- Patent, certificate:
- no
- Note:
- suitable for transfection, cells will pinocytose neutral red and will phagocytose latex beads and zymosan. They are capable of antibody-dependent lysis of sheep erythrocytes and tumor target cells. Treatment with LPS or PPD for 2 days stimulates lysis of erythrocytes, but not of tumor target cells. Easily propagated, has high DNA transfection efficiency, sensitivity to RNA interference and supports replication of murine noroviruses. Negative for surface immunoglobulin (sIg-), Ia (Ia-) and Thy-1.2 (Thy-1.2). When this line was established, it was described as not secreting detectable viral particles and negative using the XC plaque assay. Based on a published study by Dr. Janet W. Hartley in 2008, it was demonstrated that this line expresses ecotropic and polytropic MuLV and is positive using the XC plaque assay for virus replication.
- Intended use:
- these samples are intended for laboratory research only.
